The twin-arginine translocation (Tat) protein export pathway: Genetic analysis of signal peptide – translocase interactions in Escherichia coli

 

Transport of proteins across biological membranes is a crucial process in all living cells. In most bacteria, two alternative main routes exist for transporting proteins out of the cytosol: the general secretory (Sec) pathway and the so-called twin-arginine translocation (Tat) pathway. Signal peptides of the Tat pathway possess a highly conserved motif containing two consecutive arginine residues. We have analyzed signal peptide recognition by the Tat-translocase via a genetic approach using a Tat-specific, selectable reporter protein. The obtained results allowed for the mapping of signal peptide-interacting sites in the Tat translocase. Furthermore, genetic evidence will be presented for a tight cooperation between the translocase components TatB and TatC during signal peptide recognition and binding. In addition, some biotechnologically important aspects with respect to the optimization of bacterial secretory production processes for heterologous proteins will be discussed.
